Simultaneous Analytical Method for Daidzin and Glycitin in Soybean Extract

Abstract
Simultaneous analysis of daidzin and glycitin, the marker compound of soybean extract, was performed using HPLC. HPLC was analyzed with gradient elution of water/MeOH/acetic acid (88:10:2) and MeOH/acetic acid (98:2) and flow rate 1.0 mL/min, C18 MGII column (4.6 ¡¿ 250 mm, 5 ¥ìm), column temperature 40oC, PDA detector 260 nm. Daidzin and glycitin was validated for its limit of detection (LOD), limit of quantitation (LOQ), precision, accuracy and recovery by HPLC relative to soybean extract. Daidzin, glycitin standard solution and soybean extract were separated without interference from other substances, and spectrum consistent with the peak retention time appeared. It showed a high linearity in calibration curve with coefficient of correlation (R2) 0.9999. The LOD of daidzin and glycitin were 0.1211 ¥ìg/mL, 0.0493 ¥ìg/mL, LOQ of daidzin and glycitin were 0.3670 ¥ìg/mL, 0.1494 ¥ìg/mL, respectively. Inter-day precision of daidzin and glycitin were 1.3120¡¾0.0199- 1.3348¡¾0.0070 mg/g (RSD 0.5214-2.2882%) and 0.4816¡¾0.0026-0.4895¡¾0.0111 mg/g (RSD 0.5472-2.2752%). Intermediate precision of daidzin and glycitin were 1.33762¡¾0.0079 mg/g (RSD 0.5923%) and 0.4913¡¾0.0101 mg/g (RSD 2.0549%). Daidzin 0.70¡¾0.01 mg/g and glycitin 0.25¡¾0.01 mg/g were analyzed in soybean extract extracted at 90oC for 3 h. As the extraction temperature and extraction time were increased, the contents of daidzin and glycitin also increased. This result confirmed that simultaneous analysis of daidzin and glycitin of soybean extract was possible using HPLC.
